What NAD+ is

Nicotinamide adenine dinucleotide is a coenzyme present in every living cell, cycling between oxidised NAD+ and reduced NADH. Beyond its redox role it is consumed as a substrate by sirtuins, PARPs and CD38, which is what moved it from textbook biochemistry into current research interest.

Mechanism

Two distinct roles. As a redox carrier it shuttles electrons through glycolysis, the TCA cycle and oxidative phosphorylation. As a substrate it is cleaved by NAD+-consuming enzymes, linking its availability to deacetylation, DNA repair signalling and calcium signalling.

History

Discovered in 1906 by Arthur Harden and William Young while investigating yeast fermentation. Its structure was determined in the 1930s, work that contributed to Hans von Euler-Chelpin’s Nobel Prize.
